PFI-5 is an inhibitor of SMYD2 with IC50 of 8 nM. PFI-5 inhibits p53 methylation in MCF7 cells with EC50 of 1.3 uM and shows no toxicity. It also inhibits in vitro methylation of p53K370 with IC50 of 9 nM and has more than 100-fold selectivity over other histone methyltransferases and other non-epigenetic targets.
