背景信息：

干扰素基因刺激蛋白 (STING, TMEM173, MITA) 是一种跨膜接头蛋白，它是细胞病原细胞浆 DNA 的天然免疫应答的一个关键组分 (1,2)。STING 是一种被发现主要在内质网中普遍表达的蛋白 (1)。在细胞浆 DNA 的刺激下，酶 cGAMP 合成酶 (cGAS) 会产生第二信使环-GMP-AMP (cGAMP) (3,4)。cGAMP 结合并激活 STING (3,4)。此外，通过 DDX41 和 IFI16 等核酸感应蛋白检测细胞浆 DNA 会导致 STING 激活 (5,6)。STING 被激活后随 TBK1 一起转位至核周内体 (7)。TBK1 激酶磷酸化并激活干扰素调节因子 (IRF) 和 NF-κB，从而诱导 I 型干扰素和其他免疫应答基因 (1,2,7)。

激活和转运后，STING 在 Ser366（小鼠 Ser365）位点被 ULK1 磷酸化，导致 STING 失活和最终的溶酶体降解 (8)。
