BI-3406 (BI3406; SOS1-IN-2) is a novel, orally bioactive, and selective inhibitor of Son of Sevenless 1 (SOS1) with anticancer activity. It selectively binds to SOS1 and inhibits the interaction between KRAS and SOS1 with an IC50 of 6 nM. BI-3406 is able to reduce the formation of GTP-bound KRAS, thus inhibiting MAPK pathway signaling.
