体外研究

Topiramate轻微抑制分离神经元中Na+电流的持续组分，并减少Ca+和K+流阻断后，在V层锥体神经元中被诱发的Na+依赖性持久动作电位峰。[1]在大鼠杏仁基底外侧核主神经元的全细胞电压钳记录中，Topiramate在低浓度(IC50，大约为0.5 mM)下选择性抑制药理学上离体的包含GluR5亚基的红藻氨酸受体介导的兴奋性突触电流。Topiramate也显著地部分抑制AMPA受体介导的EPSCs，但是效力较低。[2] Topiramate (TPM)抑制电敏性Na+通道和非-N-甲基-D-天冬氨酸(NMDA)受体，并增强γ-氨基丁酸(GABA)介导的抑制。[3] Topiramate选择性抑制GluR5红藻氨酸受体介导的突触后响应。[4]

体内研究

Topiramate (25-100 mg/kg, i.p.)剂量依赖性提高ATPA输注诱导的阵挛发作的阈值，ATPA是一种GluR5红藻氨酸受体的选择性激动剂。[4] Topiramate剂量相关性有效抑制围产期缺氧诱发的急性癫痫发作，计算的ED50为2.1 mg/kg, i.p。[5] Topiramate (20和40 mg/kg i.p.)以剂量依赖的方式抑制癫痫强直发作和癫痫小发作，而Phenytoin (20 mg/kg i.p.)和Zonisamide (40 mg/kg i.p.)仅抑制强直发作。在DBA/2 小鼠体内，Topiramate抑制声音诱导的癫痫发作(ED50 = 8.6 mg/kg p.o.)。[6]
