一般描述

蜡状芽孢杆菌的鞘磷脂酶(Bc-Smase)是一种含Mg2+的金属酶。 蜡状芽孢杆菌 的鞘磷脂酶诱导绵羊红细胞的溶血。

Sphingomyelinase (SMase) is a hydrolase, of 34.2 kDa and its active site contain as a minimum of two magnesium atoms.

应用

Sphingomyelinase has been used in a study to assess the interaction of actin with the HIV-1 accessory protein Nef. Sphingomyelinase has also been used in a study to investigate X-ray scattering as a quality-control tool for liposomal drug-delivery systems.
Sphingomyelinase from Bacillus cereus has been used:

  • in the study to analyse the effect of sphingomyelinase on Rubella virus infectivity in cell lines
  • in lipoprotein modification for the generation of cholesterol crystals in vitro
  • to aggregate LDL (low-density lipoprotein)

生化/生理作用

Bacterial sphingomyelinase is active at neutral pH. When used in cell culture in vitro, it hydrolyzes the sphingomyelin on the outer leaflet of the plasma membrane and produces ceramide that is lipid-soluble. Sphingomyelinase is the key enzyme in the sphingomyelinase/ceramide pathway, which is implicated in the pathogenesis of several neurodegenerative disorders.
Initiates the formation of sphingomyelin-based second messengers. Activates MAPK (mitogen-activated protein kinase) and SAPKs (stress-activated protein kinases); generates ceramide from sphingomyelin.
