Lamotrigine是一种新型抗惊厥药剂，作用于人类血小板和大鼠脑突触体，抑制5-羟色胺(5-HT)吸收，IC50分别为240 μM和474 μM。

体外研究

Lamotrigine通过阻断电压依赖性钠通道稳定突触前神经元膜，从而防止兴奋性神经递质，尤其是谷氨酸和天冬氨酸的释放。[1]用10毫克/升藜芦碱培育的大鼠大脑皮层组织，lamotrigine抑制谷氨酸和天冬氨酸(两者ED 50 = 5.38 毫克/升)的释放比抑制GABA (ED50 = 11.2 毫克/升)的释放有效两倍，并且当皮层切片暴露在75毫克/升藜芦碱时，lamotrigine抑制乙酰胆碱的释放(ED50 = 25.6 毫克/升)效力降低很多。基础谷氨酸的释放不受影响。[2] Lamotrigine抑制钠依赖性动作电位的高频率持续节律放电，表明对电压激活的钠离子通道具有直接作用。[3] Lamotrigine不会诱导PCP样中枢神经系统(CNS)效应，不会通过直接抑制NMDA受体发挥作用，并可能导致与NMDA阻断相关的不良效应缺乏。[4]

体内研究

在小鼠和大鼠体内，lamotrigine防止MES和戊四氮诱导的后肢伸展，表明在动物中具有抗癫痫作用。lamotrigine给药后1小时这些作用达到最大值，并且能够持续超过24小时。[4] Lamotrigine在电诱发的EEG后放电测试中是活跃的，这表明其能够抵抗简单和复杂的部分性发作。大鼠体内lamotrigine以>5 毫克/千克的剂量静脉内给药，后放电持续时间剂量依赖性减少。[5]

Lamotrigine is an anticonvulsant drug, which is widely used in the treatment of epilepsy and bipolar disorders. It is also used in combination with other antiepileptics to treat refractory partial seizures, generalized tonic-clonic seizures and generalized toxic-chronic seizures.
Pharmaceutical secondary standards for application in quality control, provide pharma laboratories and manufacturers with a convenient and cost-effective alternative to the preparation of in-house working standards.

应用

These Secondary Standards are qualified as Certified Reference Materials. These are suitable for use in several analytical applications including but not limited to pharma release testing, pharma method development for qualitative and quantitative analyses, food and beverage quality control testing, and other calibration requirements.
Labetalol Hydrochloride may be used as a pharmaceutical reference standard for the quantification of the analyte in pharmaceutical formulations using spectrophotometric technique.
