体外研究

Trifluridine剂量依赖性抑制小鼠骨髓细胞和人大肠癌细胞移植裸鼠的增殖。Trifluridine剂量依赖性抑制集落形成的骨髓细胞。[1] Trifluridine（FTD）和2'-脱氧-5-氟尿苷（FdUrd）被掺入DNA中的不同效率，由于在TK1和DUT的底物特异性差异，导致大量的FTD掺入DNA中。与FdUrd处理的细胞相比，Trifluridine处理的细胞显示细胞核形态。[2] 在小鼠骨髓细胞和人大肠癌细胞移植的裸鼠中，Trifluridine剂量依赖性抑制增殖。[3]

体内研究

在受感染的兔子中，Trifluridine有显著少的拭子。[4] 在小鼠中，Trifluridine增加抗肿瘤活性，掺入DNA的效率比连续输注三氟胸苷更为有效。在小鼠中，Trifluridine以TPI依赖的方式逐渐积聚在肿瘤细胞的DNA中，并显著延缓肿瘤的生长并延长生存期，相比5-FU衍生物治疗。[5] 在新西兰兔眼模型中，Trifluridine显著降低病毒滴度，更少的HSV-1阳性的眼睛/总治疗期间，低级角膜炎分数，眼角膜炎/总数，和更短的时间角膜炎的分辨率。[6]

三氟胸苷已用作选择性培养基组分，用于筛选淋巴瘤细胞、人淋巴母细胞样细胞TK6和小鼠淋巴瘤细胞L5178Y中的胸苷激酶(TK)突变体。

生化/生理作用

三氟胸苷(TFT)是一种胸苷类似物，对光敏感。TFT可作为胸苷激酶底物，用于研究该酶的特异性和动力学。磷酸化TFT可掺入DNA，诱导DNA损伤，因而TFT适合用于研究DNA修复机制。TFT也可用于研究胸苷酸合成酶抑制机制和筛选胸苷激酶突变基因。它对胃肠道癌具有抗肿瘤活性，也可用于治疗疱疹病毒性角膜炎。
