体外研究

Histamine通过2型组胺受体(H2受体)抑制ROS的产生。[1] Histamine抑制突发性呼吸时单核细胞/巨噬细胞(MO)中活性氧(ROS)的产生和释放。Histamine和白介素-2 (IL-2)协同作用以激活NK细胞的细胞毒性(NKCC)。Histamine与IL-2结合通过保护免疫系统的细胞免受氧化应激和诱导天然内源性免疫细胞毒性的损害，可以提高响应率和无病生存率。[2]

体内研究

在早期酒精性肝损伤的大鼠模型中，Histamine治疗(0.5毫克/千克或5.0 毫克/千克，每天两次) 可以防止肝损伤，使血清转氨酶水平正常，并显著降低肝脏病理比例。 Histamine 的保护作用被雷尼替丁(10 毫克/千克)和H2受体拮抗剂阻断，表明histamine作用主要通过H2受体介导。[1]在雄性大鼠伏隔核中， Histamine (30 皮克/大鼠，ICV)同时增加3,4-二羟基苯丙氨酸的积累和3,4-二羟基苯丙氨酸的浓度，并且该作用不会被H2拮抗剂zolantidine影响，表明histamine通过对H1受体的作用刺激脑边缘DA神经元。[3]相比皮下注射生理盐水的大鼠，Histamine (0.5毫克/千克皮下注射)减少46%的肝脏肿瘤重量，和41%的皮下肿瘤重量。 在大鼠恶性肿瘤中，皮下histamine注射的抗肿瘤作用会被雷尼替丁(50毫克/千克，皮下注射)抑制。[4]在Sprague-Dawley大鼠体内， Histamine (1000 毫克/千克，皮下注射) 24小时后显示出急性组织损伤，在第5天和第28天注射部位显示出病理炎症。在雄性Sprague-Dawley大鼠体内，Histamine (1000毫克/千克，皮下注射)导致Cmax为 167 mM，tmax为0.5小时，t1/2为0.95，AUC 为186 mmol-h/L。[5]

Histamine dihydrochloride has been shown to activate nitric oxide synthetase and suppress or inhibit the generation of reactive oxygen species (ROS). Inhibition of ROS by histamine dihydrochloride allows activation of T cells and NK cells by IL-2. In a rat model, histamine dihydrochloride suppressed ROS generated by Kupffer cells through the H2 histamine receptor. It is a potent vasodilator and endogenous histamine receptor agonist.
