性质描述:

从水中析出者为微黄色棱状结晶一水合物（C5H4N4S·H2O，[6112-76-1]），在140℃失去结晶水。熔点313-314℃（分解）。溶于100份沸水，极微溶于冷水，几乎不溶于醇和醚。在碱笥溶液中缓慢分解，在空气及光照中颜色变深。无气味，味微苦。

生产方法:

由4，5-二氨基-6-羟基嘧啶（见1160）与甲酸环合6-羟基嘌呤（C5H4N4O），再与五硫化二磷经置换反应制得巯嘌呤。将4，5-二氨基-6-羟基嘧啶、甲酸加入反应锅中，搅拌加热溶解，升温回流4h，减压回收甲酸至尽。加入氢氧化钠溶液溶解，加活性炭脱色，过滤。滤液冷却并用冰醋酸调节pH至6，放置后过滤出结晶，即6-羟基嘌呤。将6-羟基嘌噙和五硫化二磷加入吡啶中搅拌溶解，在118℃反应4h，即生成巯嘌呤，经后处理精制而得成品，收率近60%。

用途:

该品为次黄嘌呤类干扰核合成的药物，又称抗代谢药物。阻止核酸的合成，制止癌细胞分裂繁殖，甚至因DNA合成受阻但蛋白质合成并不停止，因而发生不平衡生长，使癌细胞死亡。巯嘌呤属于周期特异性药物，主要作用于S期，对G1期也有延缓作用；对体液免疫和细胞免疫也有抑制作用。主要用于白血病。本品也是一种生化试剂。小鼠腹腔注射LD50为157mg/kg。
