产品描述 Fenofibric acid是一种降脂剂，降低低密度脂蛋白胆固醇和甘油三脂。 靶点（IC50 & Targe） MMP PPARα 体外研究 Fibric acids，降脂药的活化形式，也是PPARα的激动剂，具有提高高密度脂蛋白的效果。Fibric acids增强脂肪酸分解代谢、降低血脂水平--主要是甘油三酸酯水平。它上调了ABCA1的表达以及apoA-I所介导的HDL生成。 Fibric acids通过增强依赖于LXR的ABCA1基因的转录来发挥它对ABCA1表达的作用[1]。 体内研究 Fenofibric acid减弱OIR小鼠中循环的内皮祖细胞（EPC）的异常增多。它对EPC调动的抑制作用是依赖于PPARα的。Fenofibric acid 抑制低氧诱导的视网膜内皮祖细胞增多、减少循环中的CXCR4阳性的EPC数量，下调血清中SDF-1水平并抑制视网膜中HIF-1a和SDF-1的过表达[1]。 细胞实验 Cell lines: RAW264细胞；THP-1细胞 Concentrations: 0-200 μmol/L Incubation Time: 48 h Method: 将PPAR激活剂fenofibric acid溶解于DMSO中并加入到含0.2%BSA的培养基。RAW263细胞用PSB洗涤，与fenofibric acid在DMEM/F-12(1:1)培养基中共孵育48小时。在最后24小时的化合物处理中，加入300 mol/L dibutyryl cAMP和poA-I (10 μg/mL)。用fenofibric acid同样地处理THP-1细胞，同时加入apoA-I（in 0.2% BSA-RPMI 1640 medium）和0.1% BSA-MEM。通过酶解确定apoA-I释放到培养基中的胆固醇和含有胆碱之磷脂质。粘连细胞溶解在0.1 N NaOH，用于后续蛋白质测定。 (Only for Reference) 动物实验 Animal Models: C57BL/6J; PPARa -/- mice Formulation: DMSO Dosages: 10 mg/kg Administration: 腹膜腔内注射 (Only for Reference) 参考文献 [1] Arakawa R, et al.
