Loperamide HCl是一种阿片受体激动剂，ED50为0.15 mg/kg。

体外研究

与人的阿片受体 delta (Ki = 48 nM)和kappa (Ki = 1156 nM)相比，Loperamide表现对micro受体的高亲和力。在转染了阿片受体的人中国仓鼠卵巢细胞，Loperamide强效刺激结合[35S]鸟苷5'-O-（3-硫代）三磷酸，EC50为56 nM，并抑制forskolin刺激的cAMP累积（IC 50 =25 nM）。Loperamide在intrapaw注射后有效抑制晚期阶段福尔马林诱导退缩。[1] Loperamide是CES2的强抑制剂，Ki为1.5 muM，但它只能微弱抑制CES1A1（IC 50 =0.44 mM）。[2] 与人的阿片受体 delta (Ki = 48 nM)和kappa (Ki = 1156 nM)相比，Loperamide以浓度依赖的方式可逆阻断K+o诱发的Ca2+ i的上升，IC50为0.9 mM。Loperamide（0.1-50 mM）浓度依赖性降低IBA峰值，IC50为2.5 mM。在试验的最高浓度，Loperamide在没有任何其它药理学试剂时充分阻止IBa。在膜电位为-60毫伏时，Loperamide也衰减NMDA诱发的电流，IC50为73 mM。[3]

体内研究

在小鼠局部胫骨肿瘤（7.5-75毫克）或颈部（4毫克/千克）的毛皮下，Loperamide，阿片样激动剂无法穿越血-脑屏障，皮下注射时同时抑制热和机械痛觉过敏。[4]
