体外研究

Clemastine Fumarate作用于HL-60细胞，更有效抑制组胺诱导的Ca2+i增高，IC50为3 nM，而Chlorpheniramine或Diphenhydramine作用时，IC50分别为20 nM 和 100 nM。[1] Clemastine Fumarate浓度≥25 μM时, 作用于人红白血病细胞系K562和人B-成淋巴细胞系SB，分别显著抑制淋巴细胞的NK 和ADCC。[2] Clemastine Fumarate 抑制组胺诱导的豚鼠回肠收缩，IC50为231 nM。[3] Clemastine Fumarate作用于稳定表达HERG通道的HEK 293细胞，有效抑制HERG K+通道，这种作用存在浓度依赖性，IC50为12 nM, 可被HERG突变型Y652A或 F656A减弱。[4] Clemastine Fumarate 作用于不依赖组胺受体，但是依赖敏感性P2X7受体的HEKhP2X7 细胞，显著加强ATP诱导的Ca2+i增强，这种作用存在浓度依赖性，EC50为10 μM, 且促进LPS诱导的人巨噬细胞中释放IL-1β。[5]

体内研究

Clemastine Fumarate按5-20 mg/kg剂量处理大鼠，显著抑制酵母聚糖诱导的爪水肿和巴豆油诱导的耳肿胀，这种作用具有剂量依赖性，抑制分别为53.6%和46.8%,按20 mg/kg剂量处理, ID50分别为 18.0 mg/kg和20.5 mg/kg。[6] Clemastine Fumarate 处理小鼠，通过与细胞内信号调节激酶(ERK)调节的促炎性细胞因子的产生，如TNF-α和IL-6强相互作用，不依赖于抑制组胺H1受体，强降低对单核细胞增生李斯特菌的先天免疫反应，导致更高的死亡率。[7]
