体外研究

Diphenhydramine阻断tetrodotoxin敏感（TTX-S）和tetrodotoxin抗性（TTX-R）钠电流，K（D）分别为48 mM和86 mM。Diphenhydramine在去极化方向转移TTX-S钠电流电导-电压曲线，但对TTX-R钠电流的影响不大。Diphenhydramine使两种类型的钠电流的稳态失活曲线的超极化方向的移位。当细胞被高频去极化脉冲反复刺激，Diphenhydramine产生了深刻的使用依赖性阻断。[1] 在CCRF-CEM和Jurkat细胞系中，Diphenhydramine以剂量和时间依赖的方式诱导凋亡，而Cimetidine不能以相似浓度诱导显著效果。Diphenhydramine诱导细胞凋亡，为形态学，流式细胞仪以及细胞色素C释放到细胞质评估。在人外周血单核细胞中，Diphenhydramine抑制细胞增殖，而不诱导凋亡。[2] Diphenhydramine（500 nM）显著降低脑导水管周围灰质神经元的基准诱导，而不会对突触后电位的频率有显著效果。Diphenhydramine在高浓度时抑制脑导水管周围灰质神经元，但在低浓度下它对该基准诱导率没有影响，并阻断响应于neurotensin和tomedial视前核的刺激。[3]
