体内研究

Flutamide (Eulexin)是一种抗雄性激素药物。Flutamide-OH是flutamide的活性代谢物形式，直接结合大鼠垂体前叶雄性激素受体，Ki是55 nM。[1] Flutamide不影响小鼠乳腺癌Shionogi SC-l 15细胞在培养基中雄性激素敏感的增殖，仅表现出抗雄激素作用，而没有雄激素作用。[2] Flutamide 和leuprolide联合使用可以用于治疗前列腺癌。[3] Flutamide导致大鼠前列腺重量从319 mg 显著降低到 245 mg。联合使用flutamide 和 LHRH激动剂引发副作用，使前列腺重量降低到101mg，并且前列腺ODC活性显著下降。[4]

激酶实验

雄激素受体试验:

100μl胞浆和100 μl饱和浓度的[3H]T含有或者不含有逐渐增加浓度的无标记T，DHT，flutamide (FLU) 或flutamide-OH (FLU-OH)在0-4°C 孵育18 h。孵育结束时，加入200 μl 葡聚糖被覆活性炭(1 %活性炭，0.1%葡聚糖T-70，0.1% 凝胶，1.5 mM EDTA以及50 mM Tris (pH 7.4))在0-4°C孵育15分钟，2300 × g离心15分钟以分离自由的和结合的T。350 μl上清转移到含有10ml水溶液的计数器管中，用Beckman LS 330计数器计数。

细胞实验

Cell lines: 雄激素敏感的 Shionogi 细胞

Concentrations: ～1 μM

Incubation Time: 40天

Method: Flutamide影响培养基中小鼠乳腺癌Shionogi细胞的雄激素敏感性增殖(SEM-l)的生长。细胞在含有(MEM + 2%胎牛血清中萃取的葡聚糖被覆)和1 μM 化合物的培养基中培养40天。培养基每两天更换一次。
