PubMedPubMed 产品介绍This gene generates several transcript variants which differ in their first exons. At least three alternatively spliced variants encoding distinct proteins have been reported, two of which encode structurally related isoforms known to function as inhibitors of CDK4 kinase. The remaining transcript includes an alternate first exon located 20 Kb upstream of the remainder of the gene; this transcript contains an alternate open reading frame (ARF) that specifies a protein which is structurally unrelated to the products of the other variants. This ARF product functions as a stabilizer of the tumor suppressor protein p53 as it can interact with, and sequester, the E3 ubiquitin-protein ligase MDM2, a protein responsible for the degradation of p53. In spite of the structural and functional differences, the CDK inhibitor isoforms and the ARF product encoded by this gene, through the regulatory roles of CDK4 and p53 in cell cycle G1 progression, share a common functionality in cell cycle G1 control. WB检测(A549细胞裂解液): 样本制备:使用含1mM PMSF的RIPA裂解液冰上裂解30分钟,12000g离心15分钟取上清(p14ARF半衰期短,建议添加蛋白酶抑制剂); 电泳条件:15% SDS-PAGE凝胶(小分子量蛋白分离),恒压80V浓缩胶、120V分离胶,转膜采用0.22μm PVDF膜(200mA恒流45分钟); 抗体孵育:一抗1:1500稀释(5% BSA+TBST),4℃孵育过夜,HRP二抗(山羊抗兔)1:5000室温孵育1小时,曝光时间30-60秒(条带易被背景掩盖,建议设置阴性对照)。 IHC-P组织染色(肺癌石蜡切片): 抗原修复:采用EDTA缓冲液(pH9.0) 高压修复3分钟(针对福尔马林固定导致的表位遮蔽); 阳性定位:细胞核呈棕黄色颗粒状阳性(胞质着色为非特异性),正常肺组织肺泡上皮弱阳性,癌组织中因缺失呈阴性; 对照设置:必须包含p14ARF野生型(如HCT116) 和敲除细胞系(如U2OS) 作为阴阳对照。 IF共定位实验: 细胞处理:HeLa细胞经5Gy γ射线照射(诱导DNA损伤,激活ARF表达),4%多聚甲醛固定15分钟; 荧光标记:p14ARF抗体(Alexa Fluor 488标记,绿色)与p53抗体(Cy3标记,红色)共染,共聚焦显微镜下观察核内共定位(重叠区域呈黄色)。 公司正在出售的产品
